AC Clones after Alu-peR Amplification

Alu-peR protocols were optimized for the generation ofhu· man DN A probes from yeast strains containing yeast artificial chromosomes (YACs) with human inserts hetween 100 and 800 kb in size. The resulting DNA probes were used in chro· mosome in situ suppression (CISS) hybridization experi· ments. Strong fiuorescent signals on both chromatids indio cated the localization of specific Y AC clones, while two clearly distinguishable signals were observed in ;::.90% ot diploid nu· dei. Signal intensities were generally comparable to those ob· served using chromosome-specific alphoid DNA probes. This approach will facilitate the rapid mapping of Y AC clones and their use in chromosome analysis at aU stages of the cell eycle. © 1992 Academic Press. Ine.
